Abstract

|
Bacterial adherence occured by fimbrial adhesin is basic stage of infection. Our purpose in the experiments herein described was to cnfirm the immunological role and characterization of fimbrial adhesin (or fimbrial antigen complex:FAC) and the characteristics of the fimbrial adhesin. The degree of fimbriation in Escherichia coli was identified in electron microscope by means of negative staining with phosphotungstic acid. Purified fimbrial adhesin of Escherichia coli had been prepared by gel permeation chromatography, and the molecular weight of this was measured about 18,000 dalton with SDS-PAGE. The chromosomal and plasmid DNA fragments from Escherichia coli were transformed to competent Esherichia coli DH5 ¥á. Transformants with were detected with do-blot immunoassay by using the anti-fimbrial adhesin rabbit srum. Plasmid preparation from transformants were analyzed.
